About 90 % of solid tumours and 85 % of haematopoietic neoplasias are aneuploid. Aneuploidy is defined as an abnormal number of chromosomes that differs from multiples of the normal complete set of chromosomes. Moreover, hyperploidy is a characteristic feature of several feline fibrosarcoma cell lines that have been established at the Institute of Veterinary Pathology of the Justus-Liebig-University Giessen, Giessen, Germany. In this study we determined the chromosome number of different feline fibrosarcoma cell lines by counting immunofluorescent marked chromosomes by labelling a DNA-binding protein (Centromere Protein B) during interphase. Subsequently, we analysed and confirmed the aneuploidy in formalin-fixed paraffin-embedded sections of histologically diagnosed spontaneous feline fibrosarcomas by developing a protocol applicable to feline cell as well as by three-dimensional microscopic picture analysis and quantification. We determined a cut-off value of 76 signals per nucleus that was only exceeded in feline fibrosarcoma cell lines and tumour tissue sections taken from feline fibrosarcoma patients. The described approach could pave the road to a reliable method for the diagnosis of feline fibrosarcomas and possibly also other kinds of tumours. This work represents an ongoing effort, and it may be beneficial to integrate an algorithm capable of distinguishing between individual nuclei and effectively quantifying the signals with high accuracy, using a high-resolution scanning fluorescent microscope. By pursuing these paths, we anticipate reaching a point where we can utilize this technique more efficiently and with much less numbers of nuclei analysed to diagnose tumours by confirming the presence of aneuploidy above the threshold of 76 chromosomes, thereby corroborating the presence of malignancy.
